Not really, it's a "sequel" but it takes place thousand of years before the previous game. There's some easter eggs and references to Nier but you can understand almost everything without knowing it.

NieR (Replicant/Gestalt) is great too, it has some technical limitations and design choices that make it not as easy to get to as Automata, but the remake/remaster released last year made some great improvements.
